چکیده
A considerable body of literature exists on the biological activity of compounds containing the piperazine moiety. In the field of 1-benzyl-4-diphenylmethylpiperazines, antihistaminics,2-4) tranquillizers,5,6) and vasodilators7) have been reported. Some of them, such as buclizine (1), meclizine (2) and medibazine (3), have been used clinically. More recently, 1-diphenylmethyl-4-(4-hydroxybenzyl)piperazines (4) were disclosed to be useful in the treatment of disturbance of cerebral blood flow.8) In the course of our studies aimed at finding novel cerebral vasodilators,9) 1diphenylmethy1-4-(2,3,4-trimethoxybenzyl)piperazine (5) was found to possess considerable activity. This compound was described in the patent literature but nothing about its medical

A previous report from our laboratory described the syntheses and biological properties of 14(3-alkyl-3-hydroxy-3-pheny1)propy1]-4-(2,3,4-trimethoxybenzyppiperazine derivatives (1) as a novel class of compounds with interesting cerebral vasodilating activity.1) Encouraged
positively on the number of methoxyl groups on the benzyl moiety, and the homopiperazine analogs seem to be equipotent to the corresponding piperazines. From the standpoints of potency and ease of synthesis, 8k was selected for further study. Further analogs, which have various substituents in place of the benzyl moiety of 8k, were prepared and tested for cerebral vasodilating activity. These a...
متن کاملBenzylpiperazine derivatives. IX. Structure-antiulcer activity studies of 1-(aminocarbonylalkyl)-4-benzylpiperazine derivatives by the adaptive least-squares method.
Quantitative structure—antiulcer activity relationships of 1-(aminocarbonylalkyl)-4-benzylpiperazine derivatives (I) were analyzed by using the adaptive least-squares (ALS) technique. Discriminant functions show that (1) a bulky amide moiety is disadvantageous, (2) a small number of methylene groups between carbonyl and piperazine is favorable, (3) a substituent which has a large B1 value (or B...

Reaction of aryl methyl ketone with isatin in the presence of a base afforded 2- arylquinoline-4-carboxylic acid (3). Reaction of diazomethane with compound 3 yielded the ester 4. Compound 5 was prepared from the reaction of N, N'-dialkylethyl (or propyl) arnine with 4. Addition of hydrazine hydrate to compound 4a gave 2- (2- pyridyl) quinoline-4-carboxylic acid hydrazide (6). Reaction of l...
متن کاملSynthesis and anticancer evaluation of novel acenaphtho [1,2-e]-1,2,4- triazine derivatives
In this paper we present the convenient syntheses of seven new phenyl hydrazin derivatives 8 (a-h). For this purpose, acenaphtho [1,2-e]-1,2,4-triazine-9(8H)-thione (3) was prepared, starting from acenaphthylene-1, 2-dione (1) and thiosemicabazide in good yield. The reaction of (3) with benzyl chloride resulted to synthesis of 9-(benzylthio)-acenaphtho[1,2-e]-1,2,4-triazines (5) that reacted wi...
متن کاملSyntheses of aromatic aldehyde imine derivatives of 2-thiobenzyl-1,3,4-thiadiazole and evaluation of their anticonvulsant activity
Benzyl and chlorobenzyl substituted 1, 3, 4-thiadiazole imine derivatives have been prepared by refluxing aromatic aldehyde imine derivatives and benzyl chloride/ 4-chloro benzyl chloride in ethanolic potassium hydroxide. The structures of the compounds have been determined by spectral and chemical methods. The anticonvulsant activity has been carried out using Karl et al. method. All the repor...
